Clinical efficacy of memantine and acetylcholinesterase inhibitors as monotherapies for treatment of Alzheimer disease (AD) is well documented. Preclinical rationale and clinical evidence from large-scale, robustly designed studies make combined treatment with memantine and donepezil a natural option for the moderate stages of AD onwards. The treatment is supported by health economic analyses and acknowledged in many treatment guidelines worldwide.

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder marked in part by an accumulation of beta-amyloid, a product of aberrant proteolysis. In this Interactive Forum, the evidence supporting a pathogenic role for beta-amyloid will be explored across the spectrum of AD, from the preclinical phase through clinical dementia. Faculty will examine emerging insights into early diagnosis and amyloid-based disease-modifying strategies. |
